After Jimmy builds a DNA tester, it claims that Timmy, B.O.B., and Jimmy are all males and SpongeBob is the girl. After they replace the fuse, the machine draws a vertical line through the minus sign, completing the plus mark and proving that SpongeBob is a boy. Jenny then becomes sad, hoping SpongeBob could be her friend.

- After the Zookeeper at the zoo's states to a visiting school group that the village has three males and a female person, SpongeBob, Jimmy, Timmy and B.O.B. are confused as to which one of them is the female. After Jimmy builds a DNA tester, it claims that Timmy, B.O.B., and Jimmy are all males and SpongeBob is the girl. Jimmy attempts to hide the results, but SpongeBob pushes him out of the way to look and is very shocked. They perform another test to see if he is a girl or not. Blindfolded, SpongeBob must put a thumbtack on one of two sides of a canvas. The monster truck side is shown, but it is then revealed that he pinned the pink unicorn instead. SpongeBob starts going through five stages of grief. SpongeBob, Jimmy, Timmy and B.O.B. go on dangerous missions, but SpongeBob acts very ladylike and causes them to fail. Meanwhile, Sheen tries to make his blender work. SpongeBob then leaves the HQ to go visit Jenny and see what it's like being a girl. Jenny becomes very annoyed because she knows SpongeBob is a boy. Jenny takes SpongeBob to see Sheen, Chester and Patrick. They see that the wires gone out of control and nearly kills Sheen, Chester and Patrick. SpongeBob, still believing that he is a girl, decides that the mission is not ladylike enough. Then Timmy, Jimmy, and B.O.B. show up to save Sheen, Chester and Patrick, but end up getting stuck too. Eventually, SpongeBob decides to take off his pink bow and makes an impromptu whip/dance ribbon. After that, Timmy, Jimmy and B.O.B. take SpongeBob to do the test again and find a broken fuse. After they replace the fuse, the machine draws a vertical line through the minus sign, completing the plus mark and proving that SpongeBob is a boy. Jenny then becomes sad, hoping SpongeBob could be her friend. SpongeBob than hugs Jenny saying that the zookeeper doesn’t know what she's talking about because she's a mammal. Jenny becomes offended by this after SpongeBob says that they're all morons, and pushes SpongeBob. B.O.B. tries to hold Jenny back and says, "We'll work on that one tomorrow."
