In Los Santos, der Großstadt, in der die Spielhandlung beginnt und endet, gilt es, 100 Orte ausfindig zu machen, an denen verfeindete oder ehemals verbündete Street-Gangs ihre Kennzeichnungen – sogenannte Tags oder Graffitis – an Wänden hinterlassen haben. Die Tags sind ausschließlich in Los Santos zu finden, vermehrt in den ärmlichen Stadtteilen, vereinzelt aber auch in wohlhabenderen Gegenden. Mit Hilfe einer Sprühdose lassen sich die vorhandenen Tags der gegnerischen Banden übersprühen. Hierzu visiert man das fremde Tag an und betätigt die SCHIESSEN- bzw. O-Taste. Die Sprühdose im Haus der Johnsons ist erst nach der Mission Tagging up Turf erhältlich. Das Besprühen der Graffiti ist für die 100-Prozent-Wertung nötig. Eine Sprühdose enthält 500 Milliliter Farbe, davon braucht CJ normalerweise knapp 50 Milliliter für ein Graffiti, falls er sparsam ist. Leider kann man mit der Sprühdose keine eigenen Graffitis erstellen, die Farbe „haftet" nur auf den hundert fremden Tags. Da das Taggen verboten ist, sollte man darauf achten, dass kein Polizist in der Nähe ist, der will einen verhaften, und nach Möglichkeit sollten auch keine fremden Gangmitglieder anwesend sein, die wollen meist Schlimmeres als nur das. Marken-, Kennzeichen) beschreibt im Zusammenhang mit Graffiti das Hinterlassen eines besonderen persönlichen Schriftzugs, des sogenannten Tags, das in der Regel den grafisch gestalteten Sprühernamen enthält. I graffitin CJ sprayar, Grove Street Families står det Grove Str 4 Life i ljusgrönt. Det finns också graffiti från Seville Boulevard Families att spraya över, den är mörkgrön och det står Seville B.L.V.D Families.
