Es hija de Kurama y Hiromi, Kurama se vio completamente sorprendido cuando vio que su esposa habia dado a luz a una silpelit, tomo la drastica decision de matar a Mariko por el bien de todos, pero fue sorprendido por Hiromi, quien estaba en un estado muy fragil, hizo que Kurama prometiera perdonar la vida de su hija antes de morir, Kurama impotente sin poder romper la promesa que le habia echo Hiromi ,poco despues de su nacimiento encierra a Mariko en un cuarto de contencion evitando cualquier tipo de contacto fisico y la posibilidad de moverse,lo cual explicaria sus extremidades atrofiadas y la necesidad de moverse con la ayuda de una silla de ruedas, Saito se encarga de cuidar de ella, creyendo haber creado un lazo Madre-Hija. Una vez que el Jefe Kakuzawa ordena que utilizen a Mariko para poder capturar de nuevo a Lucy le piden a Saito que la libere,cuando ella ingresa por primera vez a la camara de contencion de Mariko, la abraza diciendole que por fin estaba con su madre, sin embargo Mariko rechazo la idea partiendo en dos a Saito enviando su torso contra el vidrio de la estacion de control, antes de que Mariko pueda matar al resto del grupo Saito logra apretar el boton que activa la bomba en su brazo, destruyendo asi su brazo e incapacitando temporalmente los vectores de Mariko.Esas bombas le fueron implantadas poco despues de nacer con el fin de poder controlarla en el futuro. Cuando es amenzada por el Jefe Kakuzawa de detonar todas las bombas en su interior, Mariko accede a colaborar con ellos y es enviada con Isobe a buscar y capturar a Lucy . Al llegar a Kamakura se enfrenta con Nana , logrando vencerla aunque perdiendo temporalmente sus vectores apartir de este punto las diferencias entre el Manga y el Anime empiezan. En el Anime, Mariko se enfrenta con Lucy dejandola fuera de combate al romper uno de sus cuernos, Kurama llega con el fin de detener a Mariko para evitar la muerte de todos, terminan reconciliandose e intentan huir juntos , pero Isobe permite que todas las bombas de Mariko sean activadas y ambos mueren. En el Manga,durante su enfrentamiento con Nana aparece Kurama para detenerla,explicandole las circunstancias de su nacimiento.Llena de odio al ver que fue rechaza por su propio padre, se propone a matar a Nana como venganza pero es detenida por Bando, Bando huye con Nana ya que Kurama se lo habia pedido, Kurama y Mariko se reconcilian, cuando Mariko salva a Kurama de un misil sin carga, el golpe hizo que Mariko sufrira un desdoblamiento emocional como Lucy/Nyu actuando de manera cariñosa con su padre.Entonces aparece Nyu en busca de sus amigos y Mariko la ataca aflorando Lucy quien tiene intenciones de matar a Kurama, Mariko se sacrifica por la vida de su padre tratando de matar a Lucy con las bombas alojadas en su cuerpo.

She was the most powerful Diclonius, with the most vectors and longest range on record. She is the daughter of Kurama and his late wife Hiromi after many unsuccessful attempts at pregnancy. During Mariko's birth, Hiromi was discovered to have cervical cancer and had to have her entire uterus removed, resulting in Mariko being their only child. Hiromi's dying plea to Kurama was for him to keep Mariko alive, but since he agreed to do whatever was demanded of him by Chief Kakuzawa without objection, he had to keep away from Mariko while she was locked away within the Diclonius Research Institute, overseen by Saito.

Mariko's father Akechi-san assassinated Lord Goroda, who was a very powerful daimyo in Japan. As a result not only was he dishonored by this action, but the Japanese considered his children to also be dishonored. Mariko had wished to be allowed to commit seppuku, but she was forced to marry Toda Buntaro, a samurai serving Lord Yoshi Toranaga. After the marriage she was sent away to Shonai Province. There was no love lost between her and the often abusive Buntaro. In revenge for leaving her alive, Mariko refused to be a properly submissive Japanese wife to him. Mariko found that she had a gift for languages and learned European languages such as English, leading to a position as an interpreter for Lord Yoshi Toranaga. When John Blackthorne arrived in Japan she was assigned as a translator between Blackthorne and the Japanese. Without Jesuits around Blackthorne was able to speak freely with Toranaga and gave him a perspective on world affairs from outside the Catholic Italian/Spanish/Portuguese perspective. As Blackthorne continued his stay on the island the two became very close friends and soon became lovers. Blackthorne petitioned Toranaga to allow Mariko to divorce Buntaro so that she would be free to marry him, but Toranaga refused to consider that and forbade Blackthorne from asking again. Mariko died when Osaka's castle was attacked by ninjas working for Ishido and the traitorus Lord Kasigi Yabu. She sacrificed herself to buy time for Blackthorne to escort hostages Ishido had been keeping to safety. Following her death Mariko was cremated in a traditional Japanese ceremony. In her will she left money to Blackthorne to enable him to build a new ship after his original ship had burned.
